  1. Methotrexate (antimetabolites)
    cancer, RA, psoriasis

    MOA: folic acid antagonist

    Toxicity- oral and GI ulcerations, myelosuppression, hepatotoxicity
  2. Anthracyclines (daunorubicin and doxorubacin)
    MOA: bind ot DNA, blockas DNA synthesis and cause breaks in DNA- generates free radicals

    Toxicity- cardiotoxic
  3. Alkylating Drugs (Cytoxan)
    • Toxicity- myelosuppression, hemorrhagic cystitis (give MESNA to prevent Hemorrhagic cystitis)
    • -Nausea/Vomit
  4. Plant alkaloids (Vincristine)
    MOA: binds to tubulin and terminates microtubule (cytoskeleton) assembly - WORKS IN METAPHASE

    Toxcitiy: alopecia and neuromuscular deficits
  5. Plant Alkaloid (vinblastine)
    • Toxcitity: leukopenia, neurological
    • blasts bone marrow (dose limiting)
  6. Alkylating drugs (Platinum containing- Cisplatin and Carboplatin)
    toxcitiy: severe nausea and vomitting, renal dysfunction, and acoustic nerve dysfunction

    most useful alkylating agent= cyclophosphamide
  7. Antiestrogens
    • Use: ER positive breast cancer treatments
    • AE: hot flashes, NV
