  1. Smear
A thin film of material containing microorganisms, spread over the surface of a slide
  2. Simple Stain
A method of staining a microorganism with a single basic dye
  3. Differential Stain
A stain that distinguishes objects on the basis of reactions to the staining procedure
  4. Gram Stain
A differential stain that classifies bacteria into two groups, gram-positive and gram-negative
  5. Gram Positive
Bacteria that retain their purple color after the decolorizer step in a Gram stain
  6. Gram Negative
Bacteria that lose their purple color after the decolorizer step in a Gram stain
  7. Micrometer
A unit of measurement equal to 10-6 meters
  8. Nanometer
A unit of measurement equal to 10-9 meters
  9. Compund Light Microscope
An instrument with two sets of lenses that uses visible light as the source of illumination
  10. Total Magnification
The magnification of a microscopic specimen, determined by multiplying the ocular lens magnification by the objective lens magnification
  11. Resolution (Resolving Power)
The ability to distinguish fine detail with a magnifying intsrument
  12. Refractive Index
The relative velocity with which light passes through a substance
  13. Flouresence Microscope
A microscope that uses an ultraviolet light source to illuminate specimens that will flouresce
  14. Electron Microscope
A microscope that uses electrons instead of light to produce an image
  15. Transmission Electron Microscope (TEM)
An electron microscope that produces high magnifications (10,000 - 100,000X) of thin sections of a specimen
  16. Scanning Electron Microscope (SEM)
An electron microscope that provides three dimensional views of the specimen magnified 1000-10,000X
  17. Staining
Colorizing a sample with a dye to view through a microscope or to visualize specific structures
  18. Fixed (Fixing)
In slide preparation, the process of attachng a specimen to a slide
  19. Acid Fast Stain
A differential stain used to identify baceria that are not decolorized by acid-alcohol
